PITTSBURGH, Pa. — Cybergenetics, the developer of TrueAllele Technology, announces the successful completion of a comprehensive forensic validation that establishes TrueAllele Casework as a scientifically reliable system for interpreting complex DNA mixtures generated through next generation sequencing (NGS).
Working with the Kern Regional Crime Laboratory, Cybergenetics validated the TrueAllele software for STR data produced by Verogen’s ForenSeq DNA Signature Prep Kit (Primer Set B) on the MiSeq FGx platform. The results demonstrated that TrueAllele consistently produced accurate, sensitive, and reproducible interpretations from mixtures containing up to five contributors.
Proven Performance for NGS Mixture Interpretation
As NGS becomes more common in forensic laboratories, the complexity of DNA interpretation has increased—especially for mixtures, degraded samples, and touch DNA. This validation showed that TrueAllele Casework was already prepared to meet these demands.
The study design included:
● Single-source assessments on sensitivity, stochastic effects, reproducibility, and 45 known reference genotypes
● Mixture evaluations involving 21 mock casework samples and a range of known mixtures with varying contributor numbers and mixture ratios
● Concordance testing comparing NGS and CE results, kit versions (Primer Set A vs. B), and plate setup methods
